The Robert Clagett Farm and Magnolia Plantation are listed on the National Register of Historic Places. By the early 19th century, a village began to form along the chief road dominant from Frederick, Maryland, to Harpers Ferry, the site of one of the two nationwide armories in the United States. The coming of the Baltimore and Ohio Railroad, Chesapeake and Ohio Canal, and the establishment of the industrialized town of Weverton to the immediate west fueled the expansion of Knoxville in the 1840s.
In 1849, the Frederick Examiner journal reported that Knoxville was expand with new businesses and homes. Many of the vernacular and Greek Revival styled buildings seen in the village today date from the 1840s and 1850s when the barns , canal, and trade along the Potomac River contributed to Knoxville's growth.
In 1851, a German Reformed congregation was established in Knoxville and a new contemporary church erected up on Cemetery Hill. Even with the interruption of the Civil War, Knoxville continued to thrive as a small-town transit hub throughout the third quarter of the 19th century.
